Procedure: To a stirred solution at 0° C. of N--t-Butoxycarbonyl-O-benzylhydroxylamine, prepared as in Example 1175A, in anhydrous THF was added portionwise a 60% dispersion of sodium hydride (1.2 equiv.) in mineral oil. Mixture stirred 30 minutes ar 0° C., and then, 1-iodobutane (1.2 equiv.) was added dropwise. Reaction stirred one hour at 0° C., and than, overnight at room temperature. Reaction concentrated in vacuo. Residue taken up in EtOAc and washed with water, 1.0M NaHCO3, 1N H3PO4, and brine. Organic layer dried with Na2SO4, filtered, and evaporated. ar 0° C. Reaction stirred one hour at 0° C. overnight at room temperature Reaction concentrated in vacuo washed with water, 1.0M NaHCO3, 1N H3PO4, and brine Organic layer dried with Na2SO4 filtered evaporated

Procedure: N-(tert-Butoxycarbonyl )-O-benzylhydroxylamine (1.43 g, 6.40 mmol) was alkylated with (6) (3.00 g, 5.68 mmol) using NaH (80%, 0.19 g, 6.33 mmol) in DMF (100 mL), following the procedure used for (4). Column chromatography on silica gel with 4:1 hexane/EtOAc yielded 3.0 g (74%) of (7) as an oil: NMR δ 1.50 (s, 9 H), 1.40-1.85 (m, 12 H), 2.20-2.50 (m, 6 H), 3.27-3.70 (m, 6 H), 4.75 (s, 6 H), 7.33 (s, 15 H). Procedure: N-(tert-Butoxycarbonyl)-O-benzylhydroxylamine (4.2 g, 18.8 mmol) was dissolved in DMF (120 mL) and cooled to 0° C. Sodium hydride (80%, 0.564 g, 18.8 mmol) was added, and the mixture was stirred at 0° C. for 30 minutes. A solution of (3) (6.0 g, 18.6 mmol) in DMF (30 mL) was slowly added to the cold solution which was then allowed to stir at room temperature for 20 minutes and at 80° C. overnight. The DMF was removed under high vacuum, and the reside was quenched with H2O (50 mL) and extracted with CH2Cl2 (3×70 mL). Organic extracts were washed with saturated sodium chloride, and the solvent was removed by rotary evaporation. Silica gel column chromatography, eluting with 2:1 hexane/EtOAc produced 7.29 g (77%) of (4) as an oil: NMR δ 1.50 (s, 9 H), 1.40-1.90 (m, 8 H), 2.30 (t, 2 H, J=6), 2.40 (t, 2 H, J=6), 3.40 (t, 2 H, J=6), 3.63 (t, 2 H, J=6), 4.77 (s, 2 H), 4.80 (s, 2 H), 7.33 (s, 10 H) . Procedure: Sodium hydride (0.48 g of 80% in mineral oil, 16 mmol) was added to a solution of tert-butyl N-benzyloxycarbamate (Compound 54 Part A, 3 g, 13 mmol) in N,N-dimethylformamide (25 mL). The reaction mixture was stirred for 20 minutes then iodomethane (1.1 mL, 18 mmol) was added dropwise with cooling in an ice bath. The resulting suspension was stirred at ambient temperature under a nitrogen atmosphere overnight. The reaction mixture was quenched with water (50 mL) then extracted with diethyl ether (150 mL). The organic layer was washed with water (5×50 mL), dried over magnesium sulfate then concentrated to provide 3.1 g of tert-butyl N-benzyloxy-N-methylcarbamate as a yellow oil. with cooling in an ice bath The resulting suspension was stirred at ambient temperature under a nitrogen atmosphere overnight The reaction mixture was quenched with water (50 mL) then extracted with diethyl ether (150 mL) The organic layer was washed with water (5×50 mL) dried over magnesium sulfate then concentrated
