Procedure: 2-Cyanofuran (4.50 g, 48.3 mmol) was dissolved in methanol (25 ml), and sodium methoxide (130 mg, 2.4 mmol) was added under ice-cooling. The mixture was stirred while the temperature is slowly raised up to room temperature for 2 hours. After the reaction was completed, acetic acid (0.16 g, 2.6 mmol) was added to neutralize the reaction solution, and the solution was concentrated under reduced pressure. Diethyl ether (100 ml) was added to the concentrated residue, and insolubles were removed by filtration. The filtrate was concentrated under reduced pressure to give the crude product of methyl 2-furancarboximidate (6.0 g) as a pale yellow oil. cooling the solution was concentrated under reduced pressure Diethyl ether (100 ml) was added to the concentrated residue, and insolubles were removed by filtration The filtrate was concentrated under reduced pressure

Procedure: To a solution of 2-furonitrile (5 g, 53.71 mmol, 1.0 eq) in dry THF under an argon atmosphere was added slowly titanium isopropoxide (18.9 ml, 64.4 mmol, 1.2 eq) at r.t. The reaction mixture was stirred for 15 min. A 1M solution of EtMgBr in THF (129 ml, 129 mmol, 2.4 eq) was added slowly via syringe to the mixture at 0° C. After addition, the reaction mixture was allowed to warm to r.t. and stirred at r.t. for 2 hour. BF3.Et2O (10.4 ml) was then added slowly to the mixture at 0° C. After the completion of addition, the reaction mixture was stirred at r.t. for 30 min. The reaction mixture was added with ice, and then with 10% NaOH solution slowly till the reaction mixture becomes basic. The mixture was extracted with EtOAc. The organic was washed with brine, dried over Na2SO4 and concentrated. The crude product was purified by silica gel (60-120) column chromatography using 35% EtOAc/Pet ether as eluent. Yield: 2.5 g (crude). After addition stirred at r.t. for 2 hour After the completion of addition the reaction mixture was stirred at r.t. for 30 min The reaction mixture was added with ice The mixture was extracted with EtOAc The organic was washed with brine dried over Na2SO4 concentrated The crude product was purified by silica gel (60-120) column chromatography

Procedure: 2-Cyanofuran (150 mg, 1.61 mmol) was dissolved in EtOH (1.61 mL) and treated dropwise with aq. hydroxylamine (50% w/w, 296 μL, 4.83 mmol) at room temperature. The mixture was warmed at 80° C. for 1.5 h. Upon completion, the solvent was removed to afford a colorless oil that was used without purification (203 mg, quant.): 1H NMR (CDCl3, 500 MHz) δ 9.34 (br s, 1H), 7.42 (d, 1H, J=1.1 Hz), 6.79 (d, 1H, J=3.3 Hz), 6.42 (dd, 1H, J=1.8, 3.4 Hz), 5.09 (br s, 2H). Upon completion, the solvent was removed to afford a colorless oil that was used without purification (203 mg, quant.)

Procedure: To a 15-mL polypropylene centrifuge tube was added 3.78 mL of 50 mM potassium phosphate buffer (pH 7.0), 1.0 mL of a suspension of 22.1 mg dry cell weight E. coli SW132 wet cells (prepared as described in Example 10) in 50 mM potassium phosphate buffer (pH 7.0), and 0.2381 g of 2-furancarbonitrile. The final concentration of 2-furancarbonitrile was 0.506 M. The reaction mixture was mixed on a rotating platform at 23° C. After 30 min, 7.50 mL of 95:5 acetonitrile/water containing 0.30 M N,N-dimethylbenzamide (HPLC external standard) was added to the reaction, the resulting mixture centrifuged, and a 0.100 mL of the supernatant mixed with 0.900 mL of acetonitrile and analyzed by HPLC. The conversion of 2-furancarbonitrile was 100%, and the yields of 2-furancarboxamide and 2-furancarboxylic acid were 99% and 0%, respectively. dry cell weight E wet cells (prepared as described in Example 10) in 50 mM potassium phosphate buffer (pH 7.0) The reaction mixture was mixed on a rotating platform at 23° C
