Procedure: 3-[4-(6-Methylsulfanyl-2-oxo-3-propyl-3,7-dihydro-2H-purin-8-yl)-bicyclo[2.2.2]oct-1-yl]-propionic acid methyl ester (105 mg) was dissolved in 2 mL of DMSO along with an appropriate amino alcohol (e.g., 160 uL of (R)-2-amino-1-butanol for compound 3). The reaction mixture was stirred at 150° C. for 3 h. It was then cooled to rt and purified by preparative HPLC to afford 50 mg of the titled compound. Procedure: 8-(4-Hydroxy-bicyclo[2.2.2]oct-1-yl)-6-methylsulfanyl-3-propyl-3,7-dihydro-purin-2-one (125 mg, 0.36 mmol) was dissolved in 3 mL of DMSO along with an excess of an appropriate amino alcohol (e.g., (R)-(−)-2-amino-1-butanol (0.24 mL, 7 eq) for compound 2). The resulting reaction mixture was stirred at 150° C. for 3 h. It was then cooled to rt and purified by preparative HPLC to afford 110 mg of the titled compound. Procedure: By the same procedure as described in the example 1, synthesis was carried out starting with 6-[1-[3-(isopropylamino)-2-pyridyl]piperazin-4-yl-carbonyl]nicotinic acid and using (R)-(−)-2-amino-1-butanol. Procedure: As described for example 208, 6-(5-methyl-3-phenyl-isoxazol-4-ylmethoxy)-nicotinic acid (200 mg, 0.65 mmol) was converted, using (R)-(+)-2-amino-1-butanol instead of (S)-(+)-2-amino-1-butanol, to the title compound (210 mg, 85%) which was obtained as a white solid. MS: m/e=380.2 [M−H]−.

Procedure: As described for example 208, 6-(5-methyl-3-phenyl-isoxazol-4-ylmethoxy)-nicotinic acid (200 mg, 0.65 mmol) was converted, using (R)-(+)-2-amino-1-butanol instead of (S)-(+)-2-amino-1-butanol, to the title compound (210 mg, 85%) which was obtained as a white solid. MS: m/e=380.2 [M−H]−.

Procedure: To an ice cold, stirred suspension of sodium hydride (60% in oil, 2.1 g, 52.0 mmol) in toluene (65 mL) was added dropwise a solution of (R)-2-aminobutan-1-ol (2.0 g, 22.0 mmol) in toluene (48 mL). After the addition was completed, the reaction mixture was warmed to room temperature and a solution of ethyl chloroacetate (3.0 g, 25.0 mmol) in toluene (12 mL) was added in a dropwise manner. The resulting mixture was then stirred at reflux for 20 h, cooled to room temperature, and solid ammonium chloride (2.7 g, 52.0 mmol) added to the reaction. The mixture was stirred for 20 min and then concentrated under reduced pressure. The crude material was purified by column chromatography (silica gel, 95:5 dichloromethane/methanol) to give (R)-5-ethylmorpholine-3-one (2.0 g, 70%) as a light yellow solid. To ice-cold tetrahydrofuran (10 mL) was added lithium aluminum hydride (1.0 M solution in tetrahydrofuran, 31.0 mL, 31.0 mmol). Once the addition was complete, a solution of (R)-5-ethylmorpholine-3-one (2.0 g, 16 mmol) in tetrahydrofuran (10 mL) was added dropwise over 20 min. Procedure: A 25 mL round-bottomed flask was charged with 2-(6-chloro-1-methyl-1H-indazol-3-yl)-5-(2-trimethylsilanyl-ethoxymethyl)-5H-pyrrolo[2,3-b]pyrazine-7-carboxylic acid (0.10 g, 0.22 mmol), (R)-2-aminobutan-1-ol (65 mg, 0.73 mmol), HBTU (108 mg, 0.28 mmol), and HOBT (44 mg, 0.28 mmol). Then added DMF (1 mL) followed by N,N-diisopropylethylamine (0.11 mL, 0.66 mmol). The yellow reaction mixture was stirred at room temperature overnight then quenched with water and extracted with EtOAc (30 mL). The organic layer was washed once with saturated NaHCO3 and water. The combined aqueous layers were back extracted with EtOAc (25 mL). The combined organic layers was dried over sodium sulfate, filtered and concentrated. The residue was purified by chromatography over SiO2 with EtOAc/hexanes (gradient: 0-50% EtOAc) to afford 70 mg (61%) of 2-(6-chloro-1-methyl-1H-indazol-3-yl)-5-(2-trimethylsilanyl-ethoxymethyl)-5H-pyrrolo[2,3-b]pyrazine-7-carboxylic acid ((R)-1-hydroxymethyl-propyl)-amide as an off-white solid. then quenched with water extracted with EtOAc (30 mL) The organic layer was washed once with saturated NaHCO3 and water The combined aqueous layers were back extracted with EtOAc (25 mL) The combined organic layers was dried over sodium sulfate filtered concentrated The residue was purified by chromatography over SiO2 with EtOAc/hexanes (gradient: 0-50% EtOAc)
