Procedure: To a suspension of 4-(phthalimidoacetyl)-2,3,3a,4,9,10a-hexahydrobenzo[b]cyclopenta[e][1,4]diazepin-10(1H)-one (389 mg, 1.0 mmol) in N,N-dimethylformamide (3 mL) was added sodium hydride (60% liquid paraffin dispersion, 44 mg, 1.1 mmol). The mixture was stirred for 10 minutes at room temperature. To this mixture was added 2-methoxybenzyl bromide (241 mg, 1.2 mmol), which was stirred for 90 minutes at room temperature. To the reaction mixture was added a saturated aqueous solution of ammonium chloride (3 mL), which was diluted with water. The resulting solid was collected by filtration, which dissolved in chloroform. The solution was dried over magnesium sulfate, which was subjected to filtration, and the filtrate was concentrated under reduced pressure. The concentrate was recrystallized from chloroform-ethanol to give 234 mg (yield 46%) of a mixture of cis-compound and trans-compound. m.p. 261°-272° C. was stirred for 90 minutes at room temperature The resulting solid was collected by filtration, which dissolved in chloroform The solution was dried over magnesium sulfate, which was subjected to filtration the filtrate was concentrated under reduced pressure The concentrate was recrystallized from chloroform-ethanol

Procedure: To a solution of 1-hydroxymethyl-2-methoxybenzene (28.7 gram, 0.207 mol) in CH2Cl2 (574 ml) at 0° C. under nitrogen was added slowly PBr3 (13.66 ml, 0.145 mol). After stirring for an additional 1.5 hours, MeOH (13.66 ml) was added and stirred for 5 minutes. To this mixture was added dropwise 10% Na2CO3 (2 ml) solution and stirred for 5 minutes. The mixture was then washed with 10% Na2 CO3 (50 ml, 2×) and brine (100 ml). It was dried over MgSO4, and filtered. The filtrate was removed under vacuum to give an oil (40 gram) of 1-bromomethyl-2-methoxy-benzene. This material was used without purification. stirred for 5 minutes stirred for 5 minutes The mixture was then washed with 10% Na2 CO3 (50 ml, 2×) and brine (100 ml) It was dried over MgSO4 filtered The filtrate was removed under vacuum

Procedure: 1-(Bromomethyl)-2-methoxybenzene (J. Indian Chem. Soc.; 28, 1951, 277; 150 mg, 0.75 mmol) was added to a suspension of the compound of Example 1 (100 mg, 0.5 mmol) and caesium carbonate (406 mg, 1.2 mmol) in acetone (4 ml), under a nitrogen atmosphere. The reaction mixture was stirred at room temperature for 18 h. The mixture was filtered and the filtrate was concentrated in vacuo. To the residue was added water (10 ml) and ethyl acetate (10 ml) and the two layers were separated. The aqueous layer was washed with a further portion of ethyl acetate (10 ml) and the combined organic layers were dried (MgSO4) and concentrated in vacuo.
