Procedure: A solution of 5.00 g of (2-amino-4-chlorophenyl)methanol, 2.73 g of cyanomethyl formate and 0.04 g of 4-N,N-dimethylaminopyridine in 50 ml of anhydrous tetrahydrofuran was stirred for 72 hours at 20° C. The solvent was then evaporated in vacuo and the residue was taken up in 50 ml of ethyl acetate. The solution was washed in succession twice with in each case 100 ml of 0.01 N hydrochloric acid and saturated sodium chloride solution, dried over sodium sulfate and then concentrated by evaporation in vacuo. The residue was purified by flash chromatography on silica gel with ethyl acetate/cyclohexane (2/1) as eluent. 1.41 g (24% of theory) of the title compound was thereby obtained in the form of slightly yellowish crystals. The solvent was then evaporated in vacuo The solution was washed in succession twice with in each case 100 ml of 0.01 N hydrochloric acid and saturated sodium chloride solution dried over sodium sulfate concentrated by evaporation in vacuo The residue was purified by flash chromatography on silica gel with ethyl acetate/cyclohexane (2/1) as eluent

Procedure: To a suspension of 5-chloro-2-hydroxymethyl aniline (8.00 g) in dichloromethane (400 ml) under nitrogen at room temperature, was added triethylamine (15.6 ml). The reaction mixture was cooled to 0° C. then phenyl acetyl chloride (14.80 ml) was added dropwise over 10 mins, and the reaction mixture was allowed to warm to room temperature and stir for 2 h. The mixture was washed with 1N HCl (2×250 ml) and brine (1×250 ml). The aqueous layers were re-extracted using CH2Cl2 (1×250 ml) and the combined organic layers were dried (MgSO4), filtered and the solvent was removed under vacuum to leave an orange solid. The solid was suspended in MeOH (400 ml), sodium hydroxide (2.20 g) was added in water (100 ml) and the reaction mixture was allowed to stir under gentle heating (50° C.) for 45 mins. The methanol was removed in vacuo, then the aqueous residue was extracted with dichloromethane (1×200 ml) and the organic layer was washed with brine (1×150 ml) and saturated sodium hydrogen carbonate solution (1×150 ml). The organic layer was dried (MgSO4), filtered and the solvent was removed under vacuum to leave a solid. Trituration with diethyl ether gave the required compound (10.00 g). to warm to room temperature The mixture was washed with 1N HCl (2×250 ml) and brine (1×250 ml) The aqueous layers were re-extracted the combined organic layers were dried (MgSO4) filtered the solvent was removed under vacuum to leave an orange solid to stir under gentle heating (50° C.) for 45 mins The methanol was removed in vacuo the aqueous residue was extracted with dichloromethane (1×200 ml) the organic layer was washed with brine (1×150 ml) and saturated sodium hydrogen carbonate solution (1×150 ml) The organic layer was dried (MgSO4) filtered the solvent was removed under vacuum to leave a solid

Procedure: (2-amino-4-chlorophenyl)methanol (10.08 g, 64.0 mmol) was suspended in chloroform (250 mL) and treated with manganese dioxide (15.87 g, 183 mmol). The mixture was stirred vigorously and heated at reflux. The oxidation was followed by TLC and found to be complete after 1 hour. The suspension was filtered through a pad of CELITE™ and evaporated to dryness under reduced pressure. Further drying under high vacuum gave a dark red solid which was used in the next step without further purification. heated at reflux The suspension was filtered through a pad of CELITE™ evaporated to dryness under reduced pressure Further drying under high vacuum gave a dark red solid which was used in the next step without further purification
