Procedure: Under nitrogen gas atmosphere, 2-cyano-3,3-bismethylthio-2-propenoic methyl ester (6 g, 29.5 mmol) was added to ethanol (40 mL) along with 3-aminopyrazole (2.6 g, 31 mmol) and the mixture was refluxed for 2.5 hours. The reaction was then cooled to room temperature and precipitate was collected by filtration under vacuum. The solid was washed with ethanol and dried under vacuum to give 7-hydroxy-5-(methylthio)pyrazolo[1,5-a]pyrimidine-6-carbonitrile in 67% yield (4.1 g). This material was used for the next step without further purification. LCMS (M+1=207) the mixture was refluxed for 2.5 hours precipitate was collected by filtration under vacuum The solid was washed with ethanol dried under vacuum

Procedure: 2-cyano-3,3-bis-methylsulfanyl-acrylic acid methyl ester Compound 1a (25.0 g, 123 mmol) was combined with 97% 2-cyano-thioacetamide (12.7 g, 123 mmol) and K2CO3 (51.0 g, 369 mmol) in DMSO (300 mL) at room temperature. The reaction was stirred for 18 hrs. The cooled reaction mixture (0° C.) was slowly acidified with 1N HCl. The resultant yellow precipitate was collected by filtration and sequentially washed with 1N HCl, water, MeOH and hexane to give 6-hydroxy-4-methylsulfanyl-2-thioxo-1,2-dihydro-pyridine-3,5-dicarbonitrile Compound 1b (24.7 g, 90% yield). 1H NMR (DMSOd6) δ 2.66 (s, 3H). 13C NMR (DMSOd6) δ 118.1 (CN), 117.3 (CN), 17.4 (SMe). MS 222 (M−). The cooled reaction mixture (0° C.) The resultant yellow precipitate was collected by filtration sequentially washed with 1N HCl, water, MeOH and hexane

Procedure: When 1,1-bis(methylthio)-2-carbomethoxy-2-cyanoethylene [prepared according to the procedure described in Chem. Ber., 95, 2861 (1962)] is reacted with 2-[(4-methyl-1H-imidazol-5-yl)methylthio]ethylamine according to the procedure of Example 6, Step A, and the resultant 1-carbomethoxy-1-cyano-2-methylthio-2-{2-[(4-methyl-1H-imidazol-5-yl)-methylthio]ethylamino}ethylene treated with propargylamine by the procedure of Example 6, Step B, there is produced 1-carbomethoxy-1-cyano-2-(2-propynylamino)-2-{2-[(4-methyl-1H-imidazol-5-yl)methylthio]ethylamino}ethylene.
