Procedure: A mixture of 2-fluoro-5-iodobenzaldehyde (500 mg, 2 mmol), cyclopentylhydrazine (273 mg, 2 mmol), and cesium carbonate (1.91 g, 5 mmol) in NMP (5 ml) was stirred at 100° C. overnight. Then KOtBu (560 mg, 5 mmol) and DMF (10 ml) were added, and the mixture was stirred at 150° C. for 5 h. After cooling to r.t., the mixture was diluted with ethyl acetate (100 ml), and washed with water (3×50 ml), and dried. Evaporation of solvent afforded batch residue, which was dissolved in acetonitrile (50 ml), and the insoluble material was removed by filtration. Flash chromatography on silica gel (n-heptane/ethyl acetate) afforded yellow oil, 158 mg (25%). the mixture was stirred at 150° C. for 5 h After cooling to r.t. washed with water (3×50 ml) dried Evaporation of solvent afforded batch residue, which the insoluble material was removed by filtration Flash chromatography on silica gel (n-heptane/ethyl acetate) afforded yellow oil, 158 mg (25%)

Procedure: Following the procedure for the preparation of 5-amino-1-(2-methoxyphenyl)-1H-pyrazole-4-carbonitrile but substituting 1-cyclopentylhydrazine and 2-(1-methoxyethylidene)malononitrile provided the title compound. 1H NMR (300 MHz, CDCl3): δ 4.24 (m, 3H), 2.24 (s, 3H), 2.01 (m, 4H), 1.90 (m, 2H), 1.67 (m, 2H).

Procedure: Alternate Synthesis of 3: Cyclopentylhydrazine hydrochloride1 (1.08 g, 10 mmol) was dissolved in 100 mL anhydrous ethanol. Sodium methoxide (540 mg, 10 mmol) was added in one portion and the reaction mixture was stirred for ten minutes. Ethoxymethylenemalononitrile (Acros, 1.22 g, 10 mmol) was then added in small portions over several minutes. The reaction mixture was heated at 70° C. under argon overnight. The reaction mixture was concentrated and subjected to flash chromatography on silica gel (stepwise elution: dichloromethane followed by 1:1 hexane: ethyl acetate) to afford 600 mg (34%) of compound 3—identical in all respects with the material obtained above. Synthesis of 3 The reaction mixture was concentrated stepwise elution

Procedure: Cyclopentylhydrazine (6.5 g, 64.9 mmol) is slowly added to a solution of ethoxymethylenemalononitrile (7.93 g, 64.9 mmol) in 100 ml of methanol at room temperature under argon, subsequently heated under reflux for 3 h and then stirred at room temperature overnight. The solvent is stripped off in a rotary evaporator and the residue is stirred with diethyl ether. The solid is filtered off with suction, washed with diethyl ether and dried under high vacuum. subsequently heated under reflux for 3 h The solvent is stripped off in a rotary evaporator the residue is stirred with diethyl ether The solid is filtered off with suction washed with diethyl ether dried under high vacuum
