Procedure: 3A Molecular sieves (10 g, 5.63 mmol) were dried under vacuum then treated with a solution of 4-methyl-6-[4-(trifluoromethyl)phenyl]pyrimidine-2-carbaldehyde (which may be prepared as described in Description 30) (1.5 g, 5.63 mmol) in dry DCM (45 mL), added under nitrogen, followed by 3-aminopyrrolidin-2-one (564.12 mg, 5.63 mmol). The mixture was gently stirred under N2 at ambient temp for 4 h. The reaction mixture was filtered through celite under suction and washed with DCM (5×20 ml). The filtrate was evaporated to an almost colourless oil which was dissolved in dry THF (60 mL) placed under nitrogen and treated with phenyl vinyl sulfone (947.7 mg, 5.6 mmol) then silver acetate (940.4 mg, 5.6 mmol). The mixture was wrapped in foil, stirred at ambient temperature for 3 minutes, then DBU (0.84 mL, 5.63 mmol) was added dropwise over a further 3 minutes and the mixture left to stir for 22 h. The reaction mixture was filtered through celite and washed through with THF (5×20 ml). The filtrate was evaporated to a dark residue, dried then dissolved in DCM (20 ml) and was applied to a 100 g silica cartridge which was initially eluted on a Biotage SP4 system with a gradient of methanol in ethyl acetate, then finally with 1M NH3-MeOH:EtOAc 1:9 to give 2-[4-methyl-6-[4-(trifluoromethyl)-phenyl]-pyrimidin-2-yl]-1,7-diazaspiro[4.4]non-1-en-6-one (D31) (774 mg); 3A Molecular sieves (10 g, 5.63 mmol) were dried under vacuum added under nitrogen The reaction mixture was filtered through celite under suction washed with DCM (5×20 ml) The filtrate was evaporated to an almost colourless oil which was dissolved in dry THF (60 mL) stirred at ambient temperature for 3 minutes the mixture left to stir for 22 h The reaction mixture was filtered through celite washed through with THF (5×20 ml) The filtrate was evaporated to a dark residue dried then dissolved in DCM (20 ml) was initially eluted on a Biotage SP4 system with a gradient of methanol in ethyl acetate

Procedure: 6′-Fluoro-N6-[4-(trifluoromethyl)pyridin-2-yl]-2,3′-bipyridine-4,6-diamine (50 mg, 0.143 mmol), 3-aminopyrrolidin-2-one (39.1 mg, 0.286 mmol), tribasic potassium phosphate (60.8 mg, 0.286 mmol), and N,N-diisopropylethylamine (0.050 ml, 0.286 mmol) were dissolved in dimethylsulfoxide (0.5 mL). The mixture was heated to 130° C. with stirring for three days. After allowing to cool to room temperature, the reaction mixture was partitioned between ethyl acetate (30 mL) and saturated aqueous sodium bicarbonate solution (15 mL). The layers were separated, and the organic layer was washed with water (3×5 mL) and brine (10 mL), dried over sodium sulfate, filtered, and concentrated under reduced pressure. The crude residue was purified using reverse phase HPLC (15 to 85% acetonitrile/water with 0.1% TFA gradient). The fractions containing the product were partitioned between 10% isopropanol/chloroform and 1:1 saturated aqueous sodium bicarbonate solution:brine. The layers were separated, and the aqueous layer was extracted with additional 10% isopropanol/chloroform. The combined organic layers were dried over sodium sulfate, filtered, and concentrated under reduced pressure to afford 3-[(4-amino-6-{[4-(trifluoromethyl)pyridin-2-yl]amino}-2,3′-bipyridin-6′-yl)amino]pyrrolidin-2-one. The reaction mixture was stirred for 16 hours at room temperature and then filtered through a pad of Celite. The filtrate was concentrated to give 3-[(E)-(4-bromo-2-pyridyl)methyleneamino]pyrrolidin-2-one as a brown gum (3.09 g, 11.53 mmol). This was dissolved in THF (120 mL) and treated with phenyl vinyl sulfone (1.94 g, 11.53 mmol), silver acetate (1.92 g, 0.59 mL, 11.53 mmol) and DBU (1.72 mL, 11.53 mmol). The reaction mixture was stirred for 3 hours at room temperature and then filtered through a pad of Celite and washed with dichloromethane. The filtrate was concentrated at reduced pressure to give a brown foam (9.53 g). This was dissolved in THF (120 mL) at 0° C. and treated with 1.7 M potassium tert-butoxide in THF (51.93 mL, 88.28 mmol) dropwise over 5 minutes. The reaction mixture was stirred at 0° C. for 1.5 hours. The reaction was quenched by the addition of acetic acid (5.04 mL, 88.28 mmol) and then filtered, washed with dichloromethane and the filtrate concentrated at reduced pressure to give a brown gum. The product was purified by silica gel chromatography eluting with 0-10% methanol in ethyl acetate to give 2-(4-bromo-2-pyridyl)-1,7-diazaspiro[4.4]non-1-en-6-one as an orange solid (830 mg); filtered through a pad of Celite The filtrate was concentrated
