Procedure: Triphenylphosphine (8.9 g, 35.2 mmol) was added to a suspension of 7-hydroxy-6-methoxy-3-((pivaloyloxy)methyl)-3,4-dihydroquinazolin-4-one (6 g, 19.6 mmol), (prepared as described for the starting material in Example 7), in methylene chloride (150 ml). This was followed by the addition of 3-methylsulphonylpropanol (3.5 g, 25.4 mmol) and diethylazodicarboxylate (5.55 ml, 35.2 mmol) in portions. The reaction was complete once the reaction became homogeneous. Silica was added and the volatiles were removed by evaporation. The free flowing powder was placed on the top of a flash chromatography column pre-equilibrated with ethyl acetate (100%). Elution was done using ethyl acetate (100%) followed by methylene chloride/ethyl acetate/methanol (60/35/3). The volatiles were removed by evaporation to give 6-methoxy-7-(3-methylsulphonylpropoxy)-3-((pivaloyloxy)methyl)-3,4-dihydroquinazolin-4-one (7.58 g, 91%) as a white solid. Procedure: 3-Methanesulfonyl-propan-1-ol (69 mg, 0.50 mmol) is dissolved in 2.0 mL of DMF and 60% sodium hydride (20 mg, 0.50 mmol) is added at 0° C. The mixture is stirred for 15 min and 3-bromo-5-bromomethyl-pyridine (50 mg, 0.20 mmol) is added. The mixture is stirred for 2 hrs and saturated aqueous NH4Cl solution (3 mL) is added along with water (10 mL) and EtOAc (10 mL). The mixture is stirred for 10 min and the aqueous layer is separated and extracted with EtOAc (2×15 ml). The organic layers are combined and concentrated to give the crude product. Purification by flash column chromatography affords 61 mg of 3-bromo-5-(3-methanesulfonyl-propoxymethyl)-pyridine. The mixture is stirred for 2 hrs The mixture is stirred for 10 min the aqueous layer is separated extracted with EtOAc (2×15 ml) concentrated to give the crude product Purification by flash column chromatography

Procedure: Tert-butyl N—[[5-(7H-pyrrolo[2,3-d]pyrimidin-4-yl)-5,8-diazaspiro[2.5]octan-8-yl]sulfonyl]carbamate (Intermediate 53)(0.12 mmol) was dissolved in dry THF (1 mL) and added 3-methylsulfonylpropan-1-ol (0.13 mmol) and triphenylphosphine (0.15 mmol). The reaction mixture was cooled to 0° C. and slowly added isopropyl (NZ)—N-isopropoxycarbonyliminocarbamate (0.15 mmol). The reaction mixture was allowed to warm up freely to rt and stirred at rt for 16 h. The crude mixture was treated with water (50 mL) and extracted with EtOAc (3×50 mL). The combined organic phases were washed with H2O (2×50 mL), brine (2×50 mL), dried over Na2SO4, filtered and concentrated in vacuo. The product was purified by flash chromatography on silica using EtOAc in heptane as eluent. to warm up freely to rt extracted with EtOAc (3×50 mL) The combined organic phases were washed with H2O (2×50 mL), brine (2×50 mL) dried over Na2SO4 filtered concentrated in vacuo The product was purified by flash chromatography on silica
