Procedure: 5-Cyano-2-hydroxyacetophenone (13.57 g) was dissolved in toluene (120 ml), and pyrrolidine (6 ml) and 1,3-dimethoxy-2-propanone (15 ml) were added. The mixture was refluxed under heating for 1 hour while removing the generated water. After cooling, the reaction mixture was diluted with ethyl acetate, washed with dilute hydrochloric acid, an aqueous solution of sodium hydrogencarbonate and brine in order, and dried over anhydrous magnesium sulfate. The solvent was distilled away and the residue obtained was purified by silica gel column chromatography (ethyl acetate:hexane=2:8) to give 13.0 g of 6-cyano-3,4-dihydro-2,2-bis(methoxymethyl)-2H-1-benzopyran-4-one. The mixture was refluxed under heating for 1 hour while removing the generated water After cooling the reaction mixture was diluted with ethyl acetate washed with dilute hydrochloric acid an aqueous solution of sodium hydrogencarbonate and brine in order, and dried over anhydrous magnesium sulfate The solvent was distilled away the residue obtained was purified by silica gel column chromatography (ethyl acetate:hexane=2:8)

Procedure: In 1,2-dichloroethane (50 ml) were suspended p-nitrobenzylamine hydrochloride (3 g), 1,3-dimethoxyacetone (1.9 g) and triethylamine (2.2 ml), and to the mixture was added, under ice-cooling, triacetoxy sodium boro hydride (4.7 g). Under nitrogen atmosphere, the mixture was stirred at room temperature for 5 hours, and to the mixture were added, under ice-cooling, 37% formalin (1.8 ml) and triacetoxy sodium boro hydride (5 g). Under nitrogen atmosphere, the mixture was stirred at room temperature overnight, and the solvent was evaporated. The residue was neutralized with 1N sodium hydroxide solution and extracted with ethyl acetate. The organic layer was washed with water and saturated brine, and dried with anhydrous magnesium sulfate. Under reduced pressure, the solvent was evaporated, and the residue was purified with silica gel column (ethyl acetate/hexane) to give N-(1,3-dimethoxypropan-2-yl)-N-methyl-4-nitrobenzylamine (3.2 g) as yellow oil. to the mixture was added, under ice- cooling to the mixture were added, under ice- cooling Under nitrogen atmosphere, the mixture was stirred at room temperature overnight the solvent was evaporated extracted with ethyl acetate The organic layer was washed with water and saturated brine dried with anhydrous magnesium sulfate Under reduced pressure, the solvent was evaporated the residue was purified with silica gel column (ethyl acetate/hexane)
