Procedure: A dry solution of 2.1 g (11.2 mmole) trans-1-amino-2-(2,2-dimethoxyethyl)-cyclohexane and 1.38 g (10.4 mmole) methoxy acetic acid isopropyl ester in 20 ml MTBE was mixed with 200 mg immobilized lipase Novozym® 435 and shaken for 25 hours at room temperature. After this period of time the conversion was 49.5%. The (1R, 2S)-enantiomer was acylated to the methoxy acetamide, while the (1S, 2R)-enantiomer was not acylated. After removal of the enzyme by filtration, removal of the solvent using an rotary evaporator and purification by column chromatography (silica gel, eluation starting with MTSE and finally with MTBE/methanol)970 mg of pure (1S, 2R)-1-amino-2-(2,2-dimethoxyethyl)-cyclohexane (46% yield) were obtained. After this period of time the conversion was After removal of the enzyme by filtration, removal of the solvent an rotary evaporator and purification by column chromatography (silica gel, eluation

Procedure: A suspension of Example B6 (0.060 g, 0.673 mmol) in DCE (4 mL) was treated with oxalyl chloride (0.059 mL, 0.673 mmol), stirred at RT for 1 h, then heated to 80° C. for 1.5 h. The mixture was cooled to RT, added drop-wise to a solution of Example A2 (0.15 g, 0.561 mmol) and pyridine (0.226 mL, 2.81 mmol) in THF (4 mL) and stirred at RT overnight. The mixture was treated with satd. Na2CO3, extracted with EtOAc (4×) and the combined organics were dried over Na2SO4 and concentrated to dryness. The material was treated with MeCN, sonicated and the resulting solid collected via filtration to afford 2-methoxy-N-((5-((2-(1-methyl-1H-pyrazol-4-yl)pyridin-4-yl)oxy)pyridin-2-yl)carbamoyl)acetamide (186 mg, 87%) as an off-white solid. 1H NMR (400 MHz, DMSO-d6): δ 10.78 (s, 1H), 10.71 (s, 1H), 8.37 (d, J=5.7 Hz, 1H), 8.27 (d, J=2.9 Hz, 1H), 8.26 (s, 1H), 8.03 (m, 1H), 7.96 (d, J=0.7 Hz, 1H), 7.74 (dd, J=9.0, 2.9 Hz, 1H), 7.23 (d, J=2.4 Hz, 1H), 6.71 (dd, J=5.7, 2.4 Hz, 1H), 4.11 (s, 2H), 3.84 (s, 3H), 3.34 (s, 3H); MS (ESI) m/z: 383.1 (M+H+). heated to 80° C. for 1.5 h The mixture was cooled to RT stirred at RT overnight The mixture was treated with satd Na2CO3, extracted with EtOAc (4×) the combined organics were dried over Na2SO4 concentrated to dryness The material was treated with MeCN sonicated the resulting solid collected via filtration

Procedure: Chlorocarbonylsulfenyl chloride (7.35 g) was added to a suspension of methoxyacetamide (5 g) in toluene (30 cm3). The reaction mixture was stirred and heated at 90°-100° C. for 5 hours, then cooled. The solvent was removed by evaporation under reduced pressure to give a brown gum (6.6 g). M+ =147; 1H NMR (DMSO-d6): δ 3.30(3H,s); 4.30(2H,s) which was used in the next step without further purification. heated at 90°-100° C. for 5 hours cooled The solvent was removed by evaporation under reduced pressure

Procedure: Intermediate 10b (562 mg, 1.50 mmol), 2-methoxyacetamide (334 mg, 3.75 mmol), Xantphos (173 mg, 0.30 mmol) and potassium carbonate (518 mg, 3.75 mmol) were suspended in 1,4-dioxane (20 mL), warmed, degassed, and placed under argon. To this mixture palladium acetate (34.0 mg, 0.15 mmol) was added. The mixture was degassed and then heated at reflux for 17 h. After cooling, the mixture was filtered through Celite, washing with DCM, and evaporated to a yellow gum. Purification by FCC, eluting with 0-100% ethyl acetate in cyclohexane, gave the title compound as a white foam (281 mg, 44%). LCMS (Method 3): Rt 3.27 min, m/z 450.2 [MNa+]. warmed degassed The mixture was degassed heated at reflux for 17 h After cooling the mixture was filtered through Celite washing with DCM evaporated to a yellow gum Purification by FCC eluting with 0-100% ethyl acetate in cyclohexane
