Procedure: To a stirred solution of 3-[2-(tert-butoxycarbonyl-pyridin-4-yl-amino)-ethoxy]-5-chloro-benzoic acid (0.100 g), TBTU (0.080 g) and HOBt (0.034 g) in DMF (3 ml) was added DIPEA (0.087 ml) followed by 3-(cyclopentylamino)propionitrile (0.035 g) after 15 min. The reaction mixture was stirred at room temperature for 18 h and then concentrated under reduced pressure. The residue was washed with water and extracted with ethyl acetate. The organic layer was dried with brine and MgSO4, filtered and concentrated to give the title compound (0.190 g) as a brown oil. concentrated under reduced pressure The residue was washed with water extracted with ethyl acetate The organic layer was dried with brine and MgSO4 filtered concentrated

Procedure: A DMF (5 mL) solution of N-(2-cyanoethyl)cyclopentylamine (4.7 mL, 32. mmol) was added to a stirred solution of HATU (15.3 g, 40.3 mmol), 1-naphthylacetic acid (5.0 g, 27 mmol), and diisopropylethylamine (7.0 mL, 40 mmol) in DMF (40 mL). The mixture stirred at room temperature overnight. Aqueous LiCl (5%) was added and the mixture was extracted three times with ethyl acetate (100 mL). The combined organic layers were washed with brine, dried over sodium sulfate, filtered, and concentrated in vacuo. The crude product was carried to next step without further purification. A sample of this product (3.35 g, 11.0 mmol), isonicotinic hydrazide (1.0 g, 7.3 mmol) and sodium methoxide (600 mg, 11 mmol) was transferred to a pressure tube. Ethanol (4 mL) was added and the reaction was stirred at 120° C. overnight. The reaction was monitored by LCMS. Upon completion, the ethanol was removed in vacuo to afford the product as a solid. The crude mixture was separated by preparative HPLC and lyophilized to afford a solid. 1H NMR (400 MHz, d6-DMSO): δ 8.8 (m, 2H), 8.2 (m, 2H), 8.0-7.85 (m, 2H), 7.8-7.7 (m, 1H), 7.6-7.3 (m, 4H), 4.5-4.3 (m, 2H), 3.5 (t, 2H), 3.3-3.0 (m, 2H), 1.8-1.3 (m, 9H). Procedure: To a cooled (0° C.) suspension of lithium aluminum hydride (3.295 g, 86.8 mmol) in anhydrous diethyl ether (150 mL) was added a solution of 3-(cyclopentylamino)propanenitrile (6.000 g, 43.4 mmol) in anhydrous diethyl ether (40 mL), dropwise over 45 mins. The reaction mixture was stirred at room temperature for 15 mins and at reflux for 4 hrs, cooled to room temperature and stirred for 1 hr. The mixture was cooled to 0° C. and vigorously stirred while water (3.4 mL) was added dropwise, followed by 15% aqueous sodium hydroxide (3.4 mL), and finally water (10.2 mL). The resultant slurry was warmed to room temperature, stirred for 15 mins, and anhydrous magnesium sulfate was added, stirring for an additional 15 mins. Solid materials were removed by filtration, washing multiple times with warm methylene chloride, and the filtrate concentrated under reduced pressure to give the desired product as a clear oil (4.5 g, 73%). 1H NMR (CDCl3, 300 MHz) δ 3.05-2.96 (quint, 1H, J=6.6 Hz), 2.74-2.71 (t, 2H, J=6.6 Hz), 2.68-2.58 (t, 2H, J=6.9 Hz), 1.85-1.68 (m, 2H), 1.62-1.42 (m, 6H), 1.34 (br s, 3H), 1.30-1.21 (m, 2H). To a cooled at reflux for 4 hrs cooled to room temperature stirred for 1 hr The mixture was cooled to 0° C. was added dropwise The resultant slurry was warmed to room temperature stirred for 15 mins stirring for an additional 15 mins Solid materials were removed by filtration washing multiple times with warm methylene chloride the filtrate concentrated under reduced pressure
