Procedure: A two-phase mixture of 4-hydroxy-N,N-dimethyl-benzenesulfonamide (13 g), 3-chloro-3-methyl-1-butyne (33 g), tetra-n-butylammonium hydrogen sulfate (22 g), and sodium hydroxide (21 g) in a mixture of dichloromethane (195 ml) and water (97.5 ml) was vigorously stirred at room temperature for 48 hours. The organic layer was separated, washed with water and brine, dried over anhydrous magnesium sulfate, and concentrated. The residue was added to n-hexane and pulverized to give N,N-dimethyl-4-(1,1-dimethyl-2-propynyloxy)benzenesulfonamide (6.4 g). Procedure: To a solution of 6.60 g (0.0330 mole) of N,N-dimethyl-4-hydroxybenzenesulfonamide dissolved in 150 ml of DMSO was added 1.32 g (0.0330 mole) of NaOH. The mixture was heated at 60° C. for 10 minutes and 7.1 g (0.030 mole) of 4-fluorophenyl phenyl sulfone was added. Procedure: A mixture of 4-hydroxy-N,N-dimethyl-benzenesulfonamide (ChemService Inc., West Chester, Pa., USA; 18 mg, 0.089 mmol), 4-(4-chloro-pyrazolo[3,4-d]pyrimidin-1-yl)-piperidine-1-carboxylic acid tert-butyl ester (Intermediate 19; 30 mg, 0.089 mmol), and potassium carbonate (27 mg, 0.196 mmol) in dimethylformamide (1 mL) was heated in a microwave oven at 160° C. for 10 min.
